Baby Signing No Need to Cave In Here is a Suggestion!

Sugestion!

I would and have done this with my 3 year old.

Say to her, "I understand you want a drink/eat right now but it is bed."

When she asks for help, you could simply say, "you are asking me to help you with something that I have already told no you can't have that. If you would like me to I can HELP you get another toy you would like to play with."

That way I am acknowledging her signed words with her and giving her other options to her.

I have found in experience that if you have given in to her, she is learning that NO doesn't really mean NO when you sign for it.
